چکیده

Recent findings demonstrate that human life is multiorganismal and microbe-host interactions play an important role in homeostasis disease. While some insights have been generated the gut, our understanding of skin still limited. We integrated metagenomic information from cutaneous microbes with transcriptome healthy, non-lesional lesional atopic psoriatic (n=272) to elucidate key pathways influencing microbial composition as well host’s transcriptome. Here, we describe pathogen-driven transcriptome-driven diversity loss dermatitis psoriasis, respectively. Patterns significantly associated these changes include expression antimicrobial peptides, compensation barrier-related genes, metal ion sequestration, alongside occurrence self-DNA lesions a potential microbe-interacting element. Among these, seven functionally heterogeneous gene clusters correlate distinct communities. Furthermore, correlated shifts microbiome were based not only on severity, but k-means clustering subjects well-established disease markers. The patterns association add up form diverse niches either succeed or fail limiting pathogen expansion. Our help compensatory mechanisms may account for fundamental differences composition.

SHP-2 Phosphatase Prevents Colonic Inflammation by Controlling Secretory Cell Differentiation and Maintaining Host-Microbiota Homeostasis

Polymorphisms in the PTPN11 gene encoding for the tyrosine phosphatase SHP-2 were described in patients with ulcerative colitis. We have recently demonstrated that mice with an intestinal epithelial cell-specific deletion of SHP-2 (SHP-2(IEC-KO) ) develop severe colitis 1 month after birth. However, the mechanisms by which SHP-2 deletion induces colonic inflammation remain to be elucidated. We ...

Cutaneous Leishmaniasis Induces a Transmissible Dysbiotic Skin Microbiota that Promotes Skin Inflammation.

Skin microbiota can impact allergic and autoimmune responses, wound healing, and anti-microbial defense. We investigated the role of skin microbiota in cutaneous leishmaniasis and found that human patients infected with Leishmania braziliensis develop dysbiotic skin microbiota, characterized by increases in the abundance of Staphylococcus and/or Streptococcus. Mice infected with L. major exhibi...

Gut Microbiota Orchestrates Energy Homeostasis during Cold

Microbial functions in the host physiology are a result of the microbiota-host co-evolution. We show that cold exposure leads to marked shift of the microbiota composition, referred to as cold microbiota. Transplantation of the cold microbiota to germ-free mice is sufficient to increase insulin sensitivity of the host and enable tolerance to cold partly by promoting the white fat browning, lead...

IRF4 promotes cutaneous dendritic cell migration to lymph nodes during homeostasis and inflammation.

Migration of resident dendritic cells (DC) from the skin to local lymph nodes (LN) triggers T cell-mediated immune responses during cutaneous infection, autoimmune disease, and vaccination. In this study, we investigated whether the development and migration of skin-resident DC were regulated by IFN regulatory factor 4 (IRF4), a transcription factor that is required for the development of CD11b...
